We are going for Umrah and landing in Madinah, then taking a train to Makkah. Can we wear the Ihrām from the hotel in Madinah (according to Sistani's rulings)?

Ehraam must be started from a Meeqaat like Masjid al Shajarah or Johfah or Qarn al-Manaazil etc. Ehraam can not start from the hotel which is not a Meeqaat. Wearing the Ehraam does not mean starting the state of Ehraam. You can wear the Ehraam in the hotel then go to the Meeqat to start you Ehraam state by Intention Niyyah of the Ehraam and reciting the Takbiyah which is :لبَّيكَداللهمَّ لبَّيك لبَّيك لا شَريكَ  لَكَ لَبَّيك إنَّ الحمدَ والنعمةَ لَكَ والمُلك، لا شريكَ لَكَ لَبَّيك  Labbaykallahumma Labbayk, Labbayka Laa Shareeka Laka Labbayk, Innal Hamda Wanni’mata Laka Walmulk, Laa Shareeka Laka Labbayk.

Is it permissible to wear Ihram from Masjid al-Shajara in Medina and travel by closed vehicles to Mecca after Isha prayer, if I am following the taqlid of Ayatollah Khoei?

According to the verdict of Ayatullah Sistani, it is not permissible for man during Ihraam to travel in a covered vehicle. If he had no choice but to do that, he has to pay Kaffara which is slaughtering a sheep or a goat and distributing it among poor believers.

Can Shia women trim their own hair and the hair of their daughters without seeking help from others after completing Saee during Umrah?

She can trim her own hair after completing Sa’ee between Safa and Marwa. She can not trim the hair of her daughter or any one else before trimming her own hair. After trimming her own hair, she will be then allowed to trim the hair of others.

Is it permissible for a Shia wife to perform Tawaaf-un-nissa on behalf of her Sunni husband who refuses to do it himself, after she has completed her own Tawaaf-un-nissa?

Sunni Muslims perform an obligatory Tawaf after Hajj and Umra , in the place of Tawaf-un-Nisa, but they call it Tawaf-ul-Widaa'. 
'Performing Tawaf-un-Nisa on behalf on someone who is unable to perform it, needs his request so that it should be done on his request. The person should be : 1. Unable to perform the worship himself for any valid reason. 2. Requesting someone to perform the worship on his behalf, for the sake of Allah (SWT).
